You'll be fine, plenty of people toss the cooler in a closet and forget about it for months at a time when aging boxes. An air tight cooler won't allow the RH to fluctuate enough to be an issue.

Without opening the cooler, where's the RH going to go? By putting the humi in there, you're wearing a belt & suspenders. The beads likely won't need refreshing until you're on the back end of your trip. And you don't have to worry about mold because there won't be any extra humidity added.

I think having the wife opening up the cooler to let air exchange introduces more problems than its worth. The less untrained hands in there, the better. Just charge those beads up the night before you leave & let them babies sit undisturbed.
